Isolation of good quality, suitably fragmented chromatin is the most important prerequisite for a successful ChIP assay. Chromatin can be sheared enzymatically or by mechanical methods such as sonication. Shearing by sonication can be advantageous for difficult to lyse cell types, It also provides random sample fragmentation preventing any bias.
